چکیده

Fifteen days after thyroidectomy, the pituitary acidophils of rats were definitely deprived of their large granules , about 350mƒÊ in diameter each, and in due succession, they displayed the changes characterized by the features of "thyroidectomy cells"(TXcells). When these rats were given a single or three injections of thyroxine, 50mg/rat, most of TX-cells might revert to their original acidophils in a short time of 6 hr. The cytomorphological course of possible reversion of the TX-cells into acidophils was clearly demonstrated on our electron micrographs. It was learned that thyroxine exhibited a strong ability to produce large granules, 350mƒÊ in diameter each, in TX-cells which might return to their original acidophils through the reduction of cell size probably due to contraction of ER and loss of intracisternal granules and through the re-production of the large granules in the ground matrix. If the rats, 15 days after thyroidectomy, were given dexamethasone (DM), 0.2mg/100g/day, for 14 days, most of TX-cells tended to slightly recover from their modifications due to thyroidectomy. DM had ability to produce both the small (130-150mƒÊ in diameter) and the large (350mƒÊ) granules in some TX-cells, but the rest of them remained unchanged and preserved a number of dilated cisternae and intracisternal granules. DM was greatly inferior to thyroxine in the ability to produce the acidophils of the large granule type (somatotrophs), while the former was superior to the latter in the ability to produce the acidophils of the small granule type. Our previous views that some TX-cells might originate from the degranulated acidophils was confirmed validly by the present observation. In conclusion, all TX-cells have not always been considered as hyperactive thyrotrophs. Mechanism of ER morphodynamics associated with vesiculation in TX-cells has been discussed with special reference to the development and reversion of TX-cells.

Fine structure of pituitary acidophils in thyroidectomized rats with or without the treatment with thyroxine and dexamethasone.

Electron microscopic study was made on rat anterior pituitaries in an attempt to clarify the causal mechanism of "loss of acidophils" which could be detected with the light microscope following thyroidectomy. In the advancing days after thyroidectomy, the acidophils (somatotrophs) were gradually deprived of their large granules whose diameter was about 350 mu. There was evidence in vivo for the...

Prolactin and luteinizing hormone cells of pregnant and lactating rats as studied by immunohistochemistry and radioimmunoassay.

The objective was to describe the cytological changes that occur in prolactin and luteinizing hormone (LH) cells of the rat hypophysis during pregnancy and lactation and to correlate these changes with secretory activity of the two cell types. The cells were demonstrated with immunohistochemistry and secretory activity was indicated by pituitary and serum hormone concentrations as measured by r...

The temporary nature of the inhibitory action of excess iodine on organic iodine synthesis in the normal thyroid.

IN 1944, Morton et al. observed that when 300 mg. of sheep thyroid slices were incubated in 3 cc. of a bicarbonate-Ringer medium to which various amounts of I as inorganic iodide had been added, inhibition of thyroxine and diiodotryosine formation occurred when the added I exceeded 20 gamma. The resemblance of this in vitro effect to the action of excess iodide in Graves' disease in man led us ...

Pectic Acid Effects on Prolactin Secretion in GH3/B6 Rat Pituitary Cell Line

Background: Pectic acid extracted from plants increases the secretion of prolactin (PRL) when injected intravenously into ewes or fed to rats. Fragments of ewe hypophysis and lactating rabbit mammary gland incubated in vitro in the presence of pectic acid secreted more PRL and caseins compared to the controls. However, it is not known whether pectic acid directly stimulates PRL secretion in pi...

Differentiation of growth hormone- and prolactin-containing acidophils with peroxidase-labeled antibody.

In order to verify the concept that growth hormone and prolactin are contained in two different populations of acidophils, sections of Bouin-fixed rat hypophyses were stained immunochemically. For this purpose the histochemical demonstration of peroxidase was utilized after sequential application to the tissue section of rabbit antiserum to human growth hormone (or antiserum to rat prolactin) f...
